In Episode 4223 of Sesame Street, the street scenes focus on Big Bird and Snuffy's playdate. Big Bird finishes up his dust bath, wanting to look nice for the playdate. When they meet, Snuffy starts sneezing violently, causing the lamppost and mailbox to hurdle away. Big Bird suspects that something nearby is making Snuffy sneeze and suggests they go to Hooper's Store instead.

At Hooper's Store, Snuffy's sneezing continues, sending a newspaper cart rolling away. Alan tries to cover Snuffy's snuffle with a tablecloth but gets blown down. They agree that the Laundromat would be a safer place to play. At the Laundromat, Leela is folding laundry when Snuffy starts sneezing again, blowing away her work table and all the clothes on it. Leela also tries covering Snuffy's snuffle but gets blown away.

Gina comes by to pick up her laundry and finds it scattered all over the place. Leela returns to work, while Gina investigates what's wrong with Snuffy. Big Bird thinks Snuffy is allergic to the laundry detergent bottle, but nothing happens when they hold it close to him. Gina notices that Snuffy starts sneezing when Big Bird gets close and conducts some experiments. She takes Big Bird far from Snuffy, who stops sneezing. After doing this several times, Big Bird realizes that Snuffy might be allergic to him. They fear they can no longer be friends, but Gina notices the dust coming off Big Bird. She deduces that it must be the dust from his dust bath that's making Snuffy sneeze. Snuffy lets out a final sneeze, blowing the dust off Big Bird, and they happily reunite.

In the "Muppets / Celebrity" segment, Taye Diggs and Idina Menzel try to explain what "allergic" means, but their animal assistants cause them to break out.

During a "Muppets" segment, Murray prepares for Abby's Flying Fairy School and plays "Bring Out Your K," asking people to display any K words they have on them. In the following "Animation" segment, the letter K is built with construction equipment.

Murray and the kids announce Abby's Flying Fairy School in another "Muppets" segment. In the "Muppets / Animation" segment, Abby's Flying Fairy School features the students cleaning up after playtime. Blogg doesn't want to put his toys away, so he sweeps them under the magic carpet. When they unlock the carpet, a hoard of Fairy Dust Bunnies is unleashed. Mrs. Sparklenose is allergic to the dust bunnies, and her sneezes cause the students to transform into various forms. The students must figure out how to return to normal and get the dust bunnies back under the rug.

In a "Muppets" segment, Elmo plays a game of "Elmo Says" with the viewer to a hip-hop beat. Murray introduces Bert and Ernie's Great Adventures in another "Muppets" segment, after finding groups of ten things in an "Animation" segment called "Ten Turtles."

In "Bert and Ernie's Great Adventures," Dustbusters Bert and Ernie are hired to rid a woman's living room of dust. While Bert prefers a broom and dustpan, Ernie opts for technology. They must cooperate to defeat a giant dust bunny.

Murray announces "Elmo's World" in a "Muppets" segment, but first, he plays "Sounds of the Street," where he and the viewer listen to the sounds of a dog, a helicopter, and a tiger with allergies. In another "Muppets" segment, Abby, Rosita, and Zoe reminisce about when Abby first arrived on Sesame Street.

A "Film / Cartoon" segment showcases the brain as the busiest part of the body. Murray and Navin introduce "Elmo's World" in a "Muppets" segment. The episode concludes with "Elmo's World: Doctors."
